Description
HATU (2-(7-Aza-1H-benzotriazol-1-yl)-N,N,N’,N’-tetramethylaminium hexafluorophosphate) is a highly efficient peptide coupling reagent widely used in both solid-phase and solution-phase peptide synthesis. It is particularly useful for forming amide bonds, especially in cases where other coupling reagents are less effective, such as with N-methyl-amino acids.

Mechanism

HATU activates carboxylic acids by forming HOAt esters, which are more reactive than OBt esters. This increased reactivity is due to the lower pKa of HOAt compared to HOBt, and the pyridine nitrogen provides anchimeric assistance to the coupling reaction, making HATU one of the most efficient coupling reagents.
